They even threw "boulders" and "large stones" through windows at the former site of Downham Special School on Horn Lane.
PC Carter said the perpetrators even threw yoghurt over the walls of the school.
Pupils are to start the new term at the Tor Bridge Campus having left the Horn Lane site at the end of term.
PC Carter, who estimated the damage from each incident would cost in region of "hundreds of pounds" to repair, added: "Fortunately the pupils aren't returning to that site otherwise it could have caused a lot of disruption when they started school.
"It's damage that's caused people inconvenience and cost them money. If you count up all the damage it creates a substantial figure.
"We've arrested two people who are now on bail pending further enquiries."
She said the dozen incidents were scattered across Plymstock and not confined to one smaller area.
At this stage the police are not looking for any body else.
