The man in his 70s was badly hurt in the blaze and was taken to Derriford Hospital, but he has died as a result of his injuries.
The blaze is believed to have started in a domestic garage connected to a bungalow in Furzehatt Park Road at 11.30am.
At the height of the fire four crews from Plympton, Plymstock, Greenbank and Crownhill were at the scene.
Firefighters say they were called after reports of smoke billowing from a domestic garage.
The road has been closed and is expected to remain closed for some time.
A police spokesman said: "The cause of the fire is not yet know and police and fire investigators will be working together to establish how the fire started."
Scenes of crime officers are at the house and are working with fire service investigators to determine the cause of the blaze.
The fire is not believed to be suspicious but a file will be prepared for the coroner.
