Gill attends UWIC in Cardiff and plays in the RFU Women's Premiership for Bristol.
Molesworth studies at Gloucestershire University and her club rugby is with Worcester.
Both are graduates of the Advanced Apprenticeship in Sports Excellence (ACE) programme at Hartpury College in Gloucestershire.
Molesworth is one of five debutants in the starting line-up for England as they try to avenge a 3-0 series defeat by France last season.
England head coach, Amanda Bennett, said: "We have selected some new, exciting players this year and some of our youngest players have really stepped up and understand what performance rugby is all about.
"We have also retained some more experienced players whose leadership is going to be much needed in the heat of battle.
"We are certainly looking forward to playing France again and start this year's campaign on home soil, so a big performance from each player is what we all want."
Meanwhile, former city rugby player Rochelle Clark and Georgia Gulliver are included in England's 23-player squad to face France in this weekend's Six Nations.
Prop Clark starts at loose-head, while scrum-half Gulliver is named on the bench.
