Brown and Drake are jointly-headlining Saturday night's show in separate contests on Chris Sanigar's five-bout card.
Promoter Sanigar is offering the duo the prize of a mouth-watering middleweight match-up early in 2009.
But only on the condition the pair pass his test – of winning in style.
Brown's opponent tomorrow night is durable Latvian Gatis Skuja, who gave another city fighter Johnny Harrison a tough work-out at the same venue last November.
Drake has been handed a new opponent and a chance to gain revenge, with Simon Fleck facing him in the opposite corner.
Leicester's Fleck, who replaces Lester Walsh, drew with Drake in the city fighter's third pro outing in June 2007 at the Aylestone Leisure Centre.
The remainder of promoter Sanigar's bill pitches Brown's Cosmo Gym stable-mate in a six-round middleweight encounter with Welshman Geraint Harvey, who replaces Rocky Chakir.
David 'Dezzy' O'Connor goes up against Reading lightweight Ibrar Riyaz, while Torquay's Ben Wakeham has a welterweight bout with Johnny Greaves.
Sanigar, fresh from steering Bristol's little big man Lee Haskins to the British superflyweight title last Friday, is expecting a high-class performance from the city duo.
Sanigar said: "Having Paul Brown and Carl Drake top any bill is always an exciting prospect.
"They're both good fighters with crowd-pleasing styles and have lots of fans in the city.
"I think a fight between the pair would be a terrific encounter and they have the chance now to prove it.
"So, it's up to these two guys now to show me and more importantly the Plymouth fight fans what they can do.
"Basically, if they want me to back them for a show with them topping the bill against each other next year, they're going to have to deliver and in style on Saturday night."
